Post-translational phosphorylation of proteins is involved in almost all important cellular processes modulating for instance protein folding, activity, interaction, localization and degradation. Therefore, incorrect phosphorylation/dephosphorylation of amino acids might have dramatic consequences for cell vitality and can be linked to cardiovascular and neurodegenerative diseases as well as cancer.
Its enormous clinical relevance renders phosphorylation analysis a challenge for biochemical and biomedical research. However, common obstacles like the transient character of phosphorylation as well as its generally low stoichiometry have to be overcome.
Due to a concerted network of kinases and phosphatases, phosphorylation patterns can alter significantly within a few seconds.
The junior group Phosphoproteomics develops novel methods and strategies for a comprehensive, quantitative and time-resolved mass spectrometric analysis of phosphorylation patterns in cells, organelles and also protein complexes – with the overall aim to improve our knowledge about cellular processes and signalling networks.
